Ocimum species are used for the improvement of anxiety and neurodegenerative disorders.

An in vitro and in vivo study was undertaken to evaluate the anti-amnesic effect of Ocimum basilicum L., O. sanctum L. and O. gratissimum L. extracts.

In vitro acetylcholinesterase (AChE) inhibitory and antioxidant activities of plant extracts were evaluated. As the most active extract was O. basilicum extract (OBE showed most marked free radical scavenging, reducing power and AChE inhibition (IC50 0.65±0.15mg/ml) activity), it was further explored for the possible anti-amnesic activity in mouse model of scopolamine induced amnesia.

The in vivo studies showed that OBE consumption (200 and 400 mg/kg) reversed the memory deficit induced by scopolamine in mice, evident by significant (p < 0.05) decrease in the transfer latency time and increase in step down latency in elevated plus maze and passive shock avoidance task, respectively. Moreover, OBE significantly reduced the brain AChE activity and oxidative stress.

Thus, the anti-amnesic and neuroprotective activities of O. basilicum may be attributed to the presence of phenolic and flavonoid compounds with antioxidant and AChE inhibitory activity.
